The problem appears to affect a fair portion of the population, leading me to think it might be an ISP/backbone related issue. It may not be possible to establish certain connections or UDP traffic may be throttled somewhere along the way to the USEast server. It could also be a hacker screwing with people/the server as is common on USEast; or it might be the server itself, going whaco for some reason.
There have been no threads made on blizzard's forum yet.
I myself am unaffected, my connection does not use NAT (my computer receives a public IP from the university routers) my computer's ports are forwarded and I'm able to join/host.
Possible fixes (for users, not for the whole problem):
Forwarding ports (6112-6119 UDP and TCP, on your computers firewall and more importantly on your router) may help, my extremely limited test group suggests those who have forwarded and can host aren't affected.
You can also "cancel" the error, one user suggested that he continually hit cancel after seeing the error until he could log in without it, after which his StarCraft worked - possibly a quick and dirty solution for those who want to play now.
