Made a melee map. Nothing special about it. Start locations aren't blocked and there is nothing really getting in the way.
When I go to SC, either Single or Multiplayer, it says there are zero players and gives the error message "You must have at least one opponent to start the game." Though there is no race selection. Take a look at the map. I'm confused as to why it doesn't work.

Post the map; we're dying to see it. If you can't upload it on SEN, just use like mediafire or something.Check the "Enable Allied Victory" box under Forces. It should show the player slots then.
You need to have other players in the lobby with you before you can start. If you set one of the players to computer, you can start by yourself.
Edit: Apparently you've already tried that.

You don't have any opponents unless another human joins the game. Change one of the spectators to a computer and your single player problem will be fixed.
Addendum: Also what Az said, you only have start locations for p1 and p2.

It didn't work when "Enable Allied Victory" was already checked. I'm troubleshooting on the phone with an SEN member who suggested unchecking it would fix. No luck.
EDIT @ Sacrieur: In single player, two human slots have the option for one to be filled by a computer player manually.

I just checked "Enable Allied Victory" and bam, two player slots showed up in-game.
Yeah I just tried that again because I was doubting it. Works now, thanks guys.

It was an error with saving. All I had to do was open the map and save it again to fix the problem.
The actual issue was map corruption, this may be related to the "read-only" error I received when I tried to save the file. But none of our suggestions were the "right" ones.
However, you do need start locations for players 3 - 8. [retracted]
