I've finally gotten around to playing through dragon age a full swing (only up to the end of the dwarf quest (my last unite all these assholes quest)) and I'm absolutely amazed by the character development in the game (with the exception of Oghren, who is an icon of everything I hate about dwarves). Anyway, here's my favorite:
Wynne: I'm not kidding, Wynne is an amazing character, and not just because I need a healer. I took the time to talk to her because of how well developed she was. Her character resonates determination to do as much as she can with what she has left of her life. I remember talking to her about her age and her response was something along the lines of "I will not sit neck deep in blankets waiting for death to take me". I also enjoy her heated debates with Morrigan, my other mage, about their religion (or rather Morrigan's lack of a religion).
**Morrigan follows as a close second, her poking at my tank Alistair is absolutely hilarious while following her personality to the letter**

As nerdy as this sounds, I honestly feel.. "close" to her when I'm playing. I don't really care about Morrigan disapproving my actions, but I don't like it when Leliana gets mad at me. D:
In terms of a character, I like Alistair the best. Sort of sarcastic, 'why me' kind of attitude, but with a sense of duty [

But, in terms of interaction between characters - has to be some of the 'philosophical' arguments that Sten and Morrigan get into. I just love his blunt way of speaking.
Morrigan. I like her art style, her coldness and the fact that she's the most useful offensive character in my party. When she isn't freezing enemies with cone of cold she's nibbing them to death with bolts from her staff.
I agree that Wynne is good as well, at keeping the party alive at least. I'm not a fan of Leliana because she's fragile and I'm already the rogue in the party - I don't want to babysit two rogues.
I purposefully started my game again as a Rogue so that I could have Sten, Morrigan and Alistair in my party. I still chuckle occasionally as we travel around Ferelden as I listen to the 3 of them bickering. Also it's a pretty powerful combo-team ... But, I like to take my time with games like this and am only about 50% complete, exploring every single rock that can be turned over, every corner of every dungeon etc...
Edit: Takes a while to get them all on the same page. Morrigan initially hates everything that makes Alistair happy, and vice versa. Sten is happy as long as you're fighting and not talking. I found it easier to keep Alistair happier initially (doing 'save the innocent villager'-type) quests, and then over time start making Morrigan happier by doing 'more evil' quests and deeds. Then when you get the Black Grimoire, boom everyone's happy. The Morale/Combat Party bonus I get is nice . Just can't find the last Revenant.

Sten didn't like me at all first time I played through. It seems the only way to make him happy is to directly oppose his own statements. I wasn't a fan of his flat personality so no biggie there.
I've only been able to get Morrigan/Leliana to 100. On my last playthrough, I got Alistair pretty high, but I ran out of things to talk to him with, and I started to run out of gifts. The only thing that I could find that makes Sten happy (besides Asala) is wine. Nothing else seems to work (or maybe I just suck at the game). Is it possible to get them all up to 100?

Sten likes paintings and art. You find lots of Watercolour Paintings etc around the game, give these to him and he'll start having more of a personality
I smacked Zevran's ass to the kerb the first time I met him. Filthy assassin. Still somehow managed to get involved with the House of Crows - or whatever they were called...
Morrigan + Alistair just for the amusing conversations.
Alistair is my tank so I got a lot of those. I also liked Alistair/Wynne's responses when I romanced Morrigan (sidenote: god damn her breasts are even bigger in the sex scene, those motherfuckers are bigger than her head!)

When I played I was mage and had Morrigan, Wynne and Sten in my party. No need for tanking with so much offensive and healing power. Seriously 2-3 chars that can resurrect fallen party members is really powerful. Anyone else got goosebumps when one day Liara sang her song at the camp fire? I couldn't help but play it again right away.
I also was thoroughly impressed at how well the characters were developed and laughed a lot early on when I moved around with Alistair and Morrigan. Favorite char: Morrigan. She's got the voice of Claudia black dudes! No but seriously she's got a very interesting personality, looks phenomenal, is a very powerful party member and her dress is really cool. I really hated Zevran though. A gay elf?! WTF?! Sten was really cool though. I don't see him as flat, he can get very philosophical, he's just usually straightforward which suits his character perfectly.
Also anyone else got the awesome bug that his armor from the main game stayed on the character WITHOUT TAKING AN ARMOR SLOT when they switched to the expansion? I started the expansion nude for some reason, but my char still had all the effects of my former armor!!!
Oh and Kaias, I agree with Samara, though you must admit that while ME2's char were still good they don't even come close to DA's chars.

I'm doing a thorough vanilla playthrough referencing the hell out of the Wiki before I mod the hell out of mine. Only thing I've modded so far was the retarded Healing Effects Received bug. I'd like to include Leliana in my party for the next playthrough, and mix up the characters a bit (Read: get the fuck out Alistair).
I actually really liked Zevran. Plus he isn't gay - he's pansexual. He's attracted to everything, which is pretty amusing.
Also, his exchanges with Morrigan (my favourite character) are funny. He's probably the only character that gets the best of her pretty consistently.
My judgements: Alistair: joker, bit moody Morrigan: funny, interesting, "my type" Leliana: annoying Sten: not mysterious enough to make his silence interesting Wynne: admirable Oghren: useless oaf Dog: a dog Zevran: have respect for his attitude Shale: voice grates on me
